Some are guarranteed employment after the leave, and some are not. In most big U.S. companies, family medical leave guarrantees return (I think there is a law about that). Personal leave of absence (a leave without a particular reason, you just say "I want to take a break") most likely won't guarrantee return.
Officially, most, if not all, types of leave of absences do not allow you to take other employment during the leave period. Otherwise the leave will be terminated automatically. Of course your company may never know what you are doing.
